Urothelium

Urothelium is a specialized epithelial tissue that lines the renal pelvis, ureters, bladder, and proximal urethra, where it forms a flexible barrier between urine and underlying tissues. Its stratified organization includes basal, intermediate, and superficial umbrella cells; umbrella cells use tight junctions, uroplakins, and regulated membrane expansion to maintain impermeability while accommodating repeated stretching during urine storage and release. Beyond its barrier role, the urothelium senses mechanical and chemical changes and communicates with nerves, smooth muscle, and immune cells. Studying urothelial structure and signaling supports research into urinary tract infections, bladder disorders, tissue injury, and regenerative medicine.

Novel Method of Plasmid DNA Delivery to Mouse Bladder Urothelium by Electroporation

We describe a novel method for the delivery of DNA plasmid into the urothelial cells of mouse bladder in vivo through urethra catheterization and electroporation. It offers a fast and convenient way for generating autochthonous mouse models of bladder diseases.
