Cell Sorting

Cell sorting is the process of separating a heterogeneous cell population into distinct groups based on physical, chemical, or biological characteristics, enabling precise analysis and experimentation. In fluorescence-activated cell sorting, cells pass individually through a flow cytometer, where lasers detect fluorescent labels or light-scattering patterns; charged droplets containing selected cells are then deflected into separate collection tubes. Magnetic-activated cell sorting instead uses antibody-coated magnetic beads to retain target cells in a magnetic field. These methods support cell isolation, immunology, stem cell research, cancer biology, and single-cell analysis by enriching rare populations and enabling functional studies of defined cell types.

Fluorescence Activated Cell Sorting of Plant Protoplasts

A method for isolating specific cell types from plant material is demonstrated. This technique employs transgenic marker lines expressing fluorescent proteins in particular cell types, cellular dissociation and Fluorescence Activated Cell Sorting. Additionally, a growth setup is established here that facilitates treatment of Arabidopsis thaliana seedlings prior to cell sorting.

In this video, we describe a protocol for the flow cytometry-based sorting of naïve B cell, memory B cell, and plasmablast plasma cell subpopulations from human peripheral blood B cells. The different B cell subpopulations can be distinguished based on fluorophore-based antibody binding to the CD19, CD27, and CD38 receptors expressed on the B cell surfaces, followed by cell sorting.

A method for producing Arabidopsis leaf protoplasts that are compatible with fluorescence activated cell sorting (FACS), allowing for studies of specific cell populations. This method is compatible with any Arabidopsis line that expresses GFP in a subset of cells.

For many scientific studies requiring a biological and chemical analysis of cell populations the cells must be in a high state of purity. Fluorescence activated cell sorting (FACS) is a superior method in which to obtain pure cell populations.
