Wolbachia Germline Transmission

Wolbachia germline transmission is the passage of Wolbachia bacteria from an infected parent to offspring through reproductive cells, most commonly from mothers through the egg cytoplasm. Because Wolbachia reside inside host cells and are generally absent from mature sperm, maternal bacteria enter developing oocytes and multiply as the germline forms, allowing infection to persist across generations. Their transmission can be enhanced by reproductive manipulations such as cytoplasmic incompatibility, in which infected males produce embryos that develop successfully mainly when eggs carry compatible Wolbachia. Studying this process helps explain host-microbe coevolution and supports applications in population biology, disease-vector control, and the spread of beneficial Wolbachia traits.

Aedes aegypti mosquitoes infected with Wolbachia are being released into natural populations to suppress the transmission of arboviruses. We describe methods to rear Ae. aegypti with Wolbachia infections in the laboratory for experiments and field release, taking precautions to minimize laboratory adaptation and selection.
