Chemical Signaling

Chemical signaling is the transmission of information between cells or organisms through chemical molecules, enabling coordination of biological activities. Signals such as hormones, neurotransmitters, and pheromones bind specific receptors on target cells, triggering intracellular pathways that alter gene expression, metabolism, movement, or behavior; signaling may occur through direct contact or across short and long distances. In biology, these processes regulate development, homeostasis, immune responses, and communication within nervous and endocrine systems. Understanding chemical signaling helps researchers explain disease mechanisms, identify drug targets, and develop therapies that modify cellular communication.

This paper uses a flow-cytometry-based assay to screen libraries of chemical inhibitors for the identification of inhibitors and their targets that influence T-cell receptor signaling. The methods described here can also be expanded for high-throughput screenings.

We describe the procedure and data analysis of a chemical screening system for glucocorticoid stress hormone signaling using zebrafish larvae: the Glucocorticoid Responsive In vivo Zebrafish Luciferase activitY (GRIZLY) assay. The assay sensitively and specifically detects effects on glucocorticoid signaling by compounds that require metabolization or affect endogenous glucocorticoid production.
