Climbing Chamber Setup

Climbing chamber setup is the preparation of a controlled vertical apparatus used to measure an organism’s ability to move upward, providing a simple assay of locomotion and behavioral performance. In a typical biology experiment, organisms are placed at the bottom of a marked chamber, allowed to climb for a defined interval, and scored by the distance reached or the proportion crossing a reference height under standardized lighting, temperature, and handling conditions. This assay supports studies of gravity-oriented movement, neuromuscular function, development, aging, and responses to drugs or environmental stress. Careful chamber design and consistent timing improve measurement reliability and enable comparisons among experimental groups.

A fast and inexpensive method for the behavioral determination of hearing parameters like hearing thresholds, hearing impairments or phantom perceptions (subjective tinnitus) is described. It uses pre-pulse inhibition of the acoustic startle response and can be easily implemented in a personal computer using a programmable AD/DA-converter and a piezo sensor.

This step-by-step protocol analyzes Drosophila negative geotaxis behavior using an automated multi-cylinder system that hosts hundreds of flies and synchronizes their action by an electric motor. Upon synchronization, fly negative geotaxis behavior is assayed, digitally recorded, and analyzed using the self-designed RflyDetection software.
