Collagen Coating

Collagen coating is a biological surface treatment that deposits collagen, a structural extracellular matrix protein, onto materials to improve their interaction with cells. The coating provides cell-adhesive sites and a tissue-like substrate, allowing cells to attach, spread, and receive extracellular signals under culture conditions. In biology, collagen-coated dishes, scaffolds, and biomaterials support the growth and differentiation of cells that respond to matrix composition, including primary cells and tissue-derived cultures. This approach is used in cell culture, tissue engineering, regenerative research, and studies of cell-matrix interactions, while coating quality and collagen organization can influence experimental reproducibility and cellular behavior.

This procedure demonstrates in vivo near IR fluorescence imaging of collagen remodeling activities in mice as well as ex vivo staining of collagens in tissue sections using caged collagen mimetic peptides that can be photo-triggered to hybridize with denatured collagen strands.

The creation of functional microtissues within microfluidic devices requires the stabilization of cell phenotypes by adapting traditional cell culture techniques to the limited spatial dimensions in microdevices. Modification of collagen allows the layer-by-layer deposition of ultrathin collagen assemblies that can stabilize primary cells, such as hepatocytes, as microfluidic tissue models.
