Conditioned Medium Preparation

Conditioned medium preparation is the process of collecting and processing cell culture medium after cells have released soluble factors into it, providing a way to study cell-to-cell signaling without direct cell contact. During incubation, cultured cells modify the medium by secreting proteins such as cytokines and growth factors, along with metabolites and extracellular vesicles; researchers then collect the medium and remove cells or debris before applying it to another culture. In biology, this approach helps assess paracrine effects on proliferation, migration, differentiation, survival, and inflammation. It supports research on stem cells, tissue repair, disease mechanisms, and the development of cell-based therapies.

This video demonstrates the production of oligodendrocyte-conditioned medium (OCM) from oligodendrocyte lineage cells by stressing them to enhance their molecular release into the medium. This medium, which is rich in growth factors, cytokines, and other bioactive molecules, can be added to neuron cultures to gain insight into the impact of oligodendrocytes-secreted factors on neuronal physiology and connectivity.

This video demonstrates an assay for promoting the growth of neuron projections known as neurite outgrowth using the macrophage-conditioned medium. Co-culture neurons and macrophages with cAMP and harvest the macrophage-derived conditioned medium. Growing the neurons with the conditioned medium promotes neurite outgrowths. These outgrowths are fluorescently labeled and visualized using a fluorescence microscope.
