Cutin Wax Polymer

Cutin-wax polymer is a hydrophobic plant biomaterial that forms the main protective barrier of the epidermal cuticle, helping aerial tissues retain water and withstand environmental stress. It develops when hydroxy fatty acids and glycerol are linked into a cross-linked polyester matrix, while soluble waxes associate with or cover this framework to reduce permeability. This layered structure also limits pathogen penetration and moderates gas and water exchange at the plant surface. Studying cutin-wax polymers helps biologists understand plant adaptation, surface development, and interactions with microbes, while informing research on crop drought tolerance, fruit storage, and biodegradable protective materials.

Here, we describe protocols for the preparation of trans-cyclobutane fused cyclooctenes (tCBCO), their polymerization to prepare depolymerizable olefinic polymers, and the depolymerization of these polymers under mild conditions. Additionally, protocols for the preparation of depolymerizable networks and compression molding of rigid linear plastics based on this system are described.

In this video, we demonstrate the processing of FFPE or formalin-fixed paraffin-embedded sections obtained from gastrointestinal tumor tissue. The deparaffinization and rehydration steps allow the processing of the tissue specimen for subsequent analytical applications.

Preparation and Friction Force Microscopy Measurements of Immiscible, Opposing Polymer Brushes

The methodology to perform friction force microscopy experiments for contacting brushes is presented: Two polymer brushes that are grafted from (a) substrates and (b) colloidal probes are slid to show that, by using two contacting immiscible brush systems, friction in sliding contacts is reduced compared to miscible brush systems.
