Cyclin A2 Function

Cyclin A2 function refers to the role of Cyclin A2, a cell-cycle regulatory protein that coordinates DNA replication and progression through cell division. Cyclin A2 accumulates during S phase and binds cyclin-dependent kinases, primarily CDK2 and CDK1, to activate phosphorylation of target proteins controlling replication, centrosome activity, and entry into mitosis; its timely degradation helps cells transition between stages. Studying Cyclin A2 function clarifies how cells regulate proliferation, development, and genome stability. Because abnormal Cyclin A2 regulation can support uncontrolled cell division, it is also relevant to cancer biology and the evaluation of cell-cycle-targeting therapies.

Metaphase to anaphase transition is triggered through anaphase-promoting complex (APC/C)-dependent ubiquitination and subsequent destruction of cyclin B. Here, we established a system which, following pulse-chase labeling, allows monitoring cyclin B proteolysis in entire cell populations and facilitates the detection of interference by the mitotic checkpoint.

Positive Regulator Molecules

2019

To consistently produce healthy cells, the cell cycle—the process that generates daughter cells—must be precisely regulated. Internal regulatory checkpoints ensure that a cell’s size, energy reserves, and DNA quality and completeness are sufficient to advance through the cell cycle. At these checkpoints, positive and negative regulators promote or inhibit a cell’s continuation through the cell cycle. Positive regulators include two protein groups that allow cells to pass through regulatory...

2019

In the present study, we describe a methodology to analyze the C924T genotype. The protocol consists of three phases: DNA extraction, amplification by polymerase chain reaction (PCR), and analysis of the restriction fragment length polymorphism (RFLP) on agarose gel.

Cyclin-dependent kinase 1 (Cdk1) is activated in the G2 phase of the cell cycle and regulates many cellular pathways. Here, we present a protocol for an in vitro kinase assay with Cdk1, which allows the identification of Cdk1-specific phosphorylation sites for establishing cellular targets of this important kinase.

Functional Groups

2020

Functional groups are a group of atoms with characteristic properties, which when linked to the carbon skeleton of a molecule, alter the properties of that molecule. For example, the presence of certain functional groups on a molecule will make them hydrophilic, whereas others will make them hydrophobic. These functional groups are an indispensable part of organic chemistry and important components of biological molecules, such as carbohydrates, proteins, lipids, and nucleic acids. Each...
