Cysteine-rich Tags

Cysteine-rich tags are engineered peptide sequences containing multiple cysteine residues, used to modify, detect, purify, or localize recombinant proteins in biological research. Their utility comes from cysteine side chains, whose reactive thiol groups can form disulfide bonds, coordinate metal ions, or react selectively with chemical probes and labeling reagents. By attaching a cysteine-rich tag to a protein, researchers can isolate it through affinity interactions, monitor its expression or cellular distribution, and study protein structure, folding, and interactions. These tags support applications in protein engineering, biochemical assays, imaging, and the development of specialized molecular tools.

A method to analyze the solvent accessibility of the thiol group of cysteine residues of Maize rayado fino virus (MRFV)-virus-like particles (VLPs) followed by a peptide cross-linking reaction is described. The method takes advantage of the availability of several chemical groups on the surface of the VLPs that can be targets for specific reactions.
