D2 Dopamine Receptors

D2 dopamine receptors are G protein-coupled receptors that detect the neurotransmitter dopamine and regulate signaling in the brain and other tissues. When activated, they primarily couple to Gi/o proteins, which inhibit adenylyl cyclase, reduce cyclic AMP production, and influence ion channels and neuronal excitability; D2 receptors can also function on presynaptic terminals to limit further dopamine release. In the basal ganglia, this signaling contributes to motor control, while in other circuits it affects motivation, learning, and cognition. D2 receptors are therefore important targets for studying Parkinson’s disease, schizophrenia, addiction, and the actions of antipsychotic and antiparkinsonian drugs.

Over the recent years, live cell-based assays have been used successfully to detect antibodies against surface and conformational antigens. Here, we describe a method using high-throughput flow cytometry enabling the analysis of large cohorts of patients. Detection of novel antibodies will improve diagnosis and treatment of immune-mediated disorders.
