Desmogleins

Desmogleins are calcium-dependent cell-adhesion proteins of the cadherin family that help neighboring cells remain mechanically connected, making them essential for tissue integrity. Located in desmosomes, their extracellular domains bind desmogleins on adjacent cells, while their intracellular tails associate with plakoglobin, plakophilins, and desmoplakin to anchor intermediate filaments. This linkage distributes mechanical stress across tissues such as the epidermis and heart. Studying desmogleins helps explain how epithelial barriers and cardiac tissue withstand strain and clarifies diseases caused by disrupted desmosomal adhesion, including pemphigus disorders, as well as broader changes in tissue organization and cancer biology.
