Cgmp Phosphodiesterase

cGMP phosphodiesterases are enzymes that terminate cyclic guanosine monophosphate (cGMP) signaling by converting cGMP into inactive GMP, thereby regulating diverse biological responses. Their catalytic domains use metal-assisted hydrolysis to break the cyclic phosphodiester bond, while distinct phosphodiesterase families and regulatory regions control where and when this activity occurs. In biology, cGMP phosphodiesterases help govern visual signal processing in photoreceptors, smooth-muscle relaxation, and cellular responses to hormones and neurotransmitters. Because altered cGMP breakdown can disrupt signaling, these enzymes are important subjects in physiology, disease research, and drug development, including therapies that target PDE5 to modulate vascular function.

Treatment for Pulmonary Arterial Hypertension: Phosphodiesterase Inhibitors

2024

Phosphodiesterase 5 (PDE5) inhibitors are potent enzymes that function to hydrolyze cyclic nucleotides to their corresponding 5' monophosphates. Their unique biochemical properties have been applied in treating Pulmonary Arterial Hypertension (PAH). Among the PDE5 inhibitors, sildenafil (Revatio) stands out as a competitive and selective inhibitor. It operates by elevating cellular levels of cGMP and augmenting signaling through the cGMP-PKG pathway, promoting vasodilation. Upon oral...

2025

This protocol measures cyclic guanosine monophosphate (cGMP) concentrations in solid tissues using competitive enzyme-linked immunosorbent assay (ELISA). The protocol describes how to process solid tissue, perform competitive ELISA, convert absorbances to cGMP concentrations in picomoles per milliliter, and calculate the final concentrations as nanomoles of cGMP per gram of tissue.

This protocol describes a novel mechanical chopping method that allows the expansion of spherical neural stem and progenitor cell aggregates without dissociation to a single cell suspension. Maintaining cell/cell contact allows rapid and stable growth for over 40 passages.

In Vitro Mouse Bone Marrow Stromal Cell Differentiation into Adipocytes

2025

This video demonstrates the differentiation of bone marrow stromal cells (BMSCs) into adipocytes. In the two-step process, in the presence of specific growth factors and hormones, undifferentiated BMSCs first differentiate into preadipocytes. The second step involving treatment with glucose and insulin leads to the formation of mature adipocytes with multiple lipid droplets.

Isolating and Culturing Murine Cerebellar Granular Neuron Progenitors

2025

This video demonstrates the isolation and cultivation of cerebellar neural progenitor cells from a mouse pup's cerebellum. Cerebellar granule neuron progenitors are isolated and purified through differential adhesion, effectively removing astrocytes. These cultured progenitors subsequently differentiate into cerebellar granule neurons.
