Differential Stain

Differential staining is a microscopy method that uses more than one stain or staining step to distinguish cells, cell structures, or microorganisms according to their chemical or physical properties. In a typical procedure, a primary dye colors the specimen, while a mordant, decolorizer, or counterstain alters dye retention; differences in cell-wall composition, permeability, or resistance to decolorization then produce contrasting colors. In biology, differential stains such as Gram and acid-fast methods help classify bacteria, assess cellular organization, and guide identification in clinical, environmental, and research samples. By making otherwise similar specimens visually distinct, the technique supports rapid analysis and complements culture and molecular methods.

In this video, we describe a technique of hematoxylin and eosin staining of paraffin-embedded tissue sections to study their histology. The procedure includes deparaffinization of the tissue followed by staining the cells in the tissue. Upon staining, the nucleus appears distinctly blue, and the proteins in the cytoplasm display pink coloration.
