Dii Staining

DiI staining is a fluorescence-based method for labeling cell membranes and tracing cellular structures in biological samples. The lipophilic dye inserts into lipid bilayers, where it diffuses laterally along the membrane while its fluorescent signal allows labeled cells and their processes to be visualized by microscopy. Researchers use DiI staining to examine cell morphology, membrane organization, neuronal projections, and developmental cell movements in tissues and organisms. Because the label can reveal connections and migration patterns over time, the technique supports studies of tissue development, neural circuitry, and cell-to-cell organization.

Visualization of Caenorhabditis elegans Cuticular Structures Using the Lipophilic Vital Dye DiI

We present a method to visualize cuticle in live C. elegans using the red fluorescent lipophilic dye DiI (1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate), which is commonly used in C. elegans to visualize environmentally exposed neurons. With this optimized protocol, alae and annular cuticular structures are stained by DiI and observed using compound microscopy.

Dual Labeling of Neural Crest Cells and Blood Vessels Within Chicken Embryos Using ChickGFP Neural Tube Grafting and Carbocyanine Dye DiI Injection

Here we report dual labeling of neural crest cells and blood vessels using chickGFP neural tube intraspecies grafting combined with intra-vascular DiI injection. This experimental technique allows us to simultaneously visualize and study development of the NCC-derived (enteric) nervous system and the vascular system, during organogenesis.

Using a lipophilic 1,1'-Dioctadecy-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI) staining technique, Ambystoma mexicanum can undergo vascular perfusion to allow for easy visualization of the vasculature.
