Disc Isolation

Disc isolation is a biological dissection technique used to separate an intact disc-shaped tissue or organ from surrounding structures for controlled study. In intervertebral disc research, careful removal of adjacent vertebral and connective tissues preserves the disc’s main components, including the annulus fibrosus and nucleus pulposus, while minimizing mechanical or structural damage. Isolated discs can then support organ culture, histological analysis, molecular assays, and investigations of tissue degeneration, biomechanics, and cellular responses. By providing a defined experimental preparation, disc isolation helps researchers examine disc development and disease mechanisms and evaluate potential therapeutic strategies under controlled laboratory conditions.

Generation, Purification, and Characterization of Cell-invasive DISC1 Protein Species

The generation, purification and cell invasion of intracellular, cytoplasmic full length DISC1 protein aggresomes from cell cultures and of a labeled, multimeric recombinant DISC1 protein fragment in E. coli are described. Cell invasiveness is shown for recipient cells in cell culture and for neurons in vivo after stereotactical brain inoculation.

We describe the isolation of cardiac extracellular matrix from C57Bl/6J control mice, tight-skin mice, and tight-skin mice treated with the IRF5 inhibitory peptide. We also describe the vasodilation studies on the isolated vessels from C57Bl/6J, tight-skin mice and tight-skin mice treated with the IRF5 inhibitory peptide.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.
