Dsrna Signaling Transfer

dsRNA signaling transfer is the movement of double-stranded RNA or its derived signals between cells, allowing genetic information and immune cues to influence neighboring or distant tissues. Cells can release dsRNA in extracellular vesicles or other carriers, while recipient cells internalize it and process it through pathways involving Dicer, small interfering RNAs, and the RNA-induced silencing complex; dsRNA can also activate innate immune sensors. This process contributes to antiviral defense, gene regulation, and communication within multicellular organisms. Studying dsRNA transfer supports research on RNA interference, host-pathogen interactions, biological regulation, and the development of RNA-based therapeutic strategies.

This video protocol illustrates a method used by the James lab to inject dsRNA into female A. aegypti mosquitoes, which harbor the dengue virus. RNA interference has been proven very effective to analyze gene function in Drosophila tracheal development. A detailed protocol used by Jiang lab to inject dsRNA into fly embryos to knockdown gene expression is illustrated. This protocol describes a microinjection methodology that we have standardized and used for several years to deliver specific quantities of nucleic acids directly to the hemolymph of mosquitoes and house flies. This protocol results in minimal injection mortality and allows dose correlated measurements of fecundity.
