E Cadherin Fibronectin

E-cadherin and fibronectin are complementary regulators of cell adhesion, linking epithelial cell organization with interactions between cells and the extracellular matrix. E-cadherin forms calcium-dependent adherens junctions through homophilic binding between neighboring cells, whereas fibronectin is an extracellular matrix glycoprotein that binds integrins and supports cell attachment, spreading, and migration. During epithelial-to-mesenchymal transition, E-cadherin levels often decrease while fibronectin production increases, weakening cell-cell cohesion and promoting matrix-associated movement. Their coordinated analysis helps researchers study tissue development, wound healing, cancer invasion, and the molecular changes that control cell phenotype and behavior.

Structure of Cadherins

2023

The cadherins were one of the first cell adhesion molecules discovered; the term “cadherins” is based on their calcium-dependent adhering properties. The first cadherins discovered on the epithelial, neuronal, and placental cells were named E-cadherin, P-cadherin, and N-cadherin, respectively. These classical cadherins share sequence and structural similarities. Other cadherins, including those involved in cell signaling, are grouped into non-classical cadherins. This diversity of cadherins...

Cadherins in Tissue Organization

The cadherins are a superfamily of cell adhesion molecules comprising over 180 variants, with specific tissues expressing a particular combination of cadherin types. Cadherins generally exhibit homophilic binding; i.e., cadherins on one cell bind to cadherins of the same or closely related type on another cell. Thus, cells of the same type have a specific affinity to bind to each other and sort themselves into clusters to form tissues. Cell Sorting During Development Cell sorting plays an...

Fibronectins Connect Cells with ECM

Fibronectin is an adhesive glycoprotein present in the extracellular matrix of embryogenic and adult tissue. These molecules primarily aid in regulating cell motility and attachment. A fibronectin molecule is composed of two identical polypeptide chains attached to each other by a pair of disulfide bonds at the C-terminal. Both proteoglycans and collagen are attached to fibronectin proteins, which, in turn, are attached to integrin proteins. These integrin proteins interact with transmembrane...

This method is useful for quantifying the early dynamics of cellular adhesion and spreading of anchorage-dependent cells onto the fibronectin. Furthermore, this assay can be used to investigate the effects of altered redox homeostasis on cell spreading and/or cell adhesion-related intracellular signaling pathways.
