E. Coli Expression

E. coli expression is a widely used biological system for producing recombinant proteins by directing genetically engineered Escherichia coli cells to transcribe and translate a target gene. Typically, researchers place a coding sequence in a plasmid downstream of a regulated promoter, introduce the plasmid into bacteria, and induce expression under controlled growth conditions; host RNA polymerase transcribes the gene, and ribosomes translate the messenger RNA into protein. The resulting proteins can be harvested and purified for biochemical assays, structural studies, enzyme production, and biotechnology research. Because E. coli grows rapidly and is easy to engineer, it supports efficient gene-function testing and scalable recombinant production, although protein folding and post-translational modification may require additional strategies.

Protein co-expression is a powerful alternative to the reconstitution in vitro of protein complexes, and is of help in performing biochemical and genetic tests in vivo. Here we report on the use of protein co-expression in Escherichia coli to obtain protein complexes, and to tune the mutation frequency of cells.
