Egfr Crosstalk

EGFR crosstalk is the bidirectional communication between the epidermal growth factor receptor and other cell-surface receptors or intracellular signaling pathways, allowing cells to integrate multiple environmental cues. When a ligand activates EGFR, receptor phosphorylation initiates signaling through networks such as RAS–RAF–MEK–ERK and PI3K–AKT, while other receptors can modify EGFR activity through transactivation, shared effectors, or feedback loops. This coordination regulates cell proliferation, survival, migration, and differentiation in normal biology. In cancer research, understanding EGFR crosstalk helps explain therapeutic resistance, identify cooperating pathways, and guide combinations of targeted treatments.

The genetically tractable nematode Caenorhabditis elegans can be used as a simple and inexpensive model for drug discovery. Described here is a protocol to identify anticancer therapeutics that inhibit the downstream signaling of RAS and EGFR proteins.

Establishing Dual Resistance to EGFR-TKI and MET-TKI in Lung Adenocarcinoma Cells In Vitro with a 2-step Dose-escalation Procedure

An in vitro method for establishing dual resistance to an EGFR-TKI and a MET-TKI in cancer cells is described. This method is useful for developing treatments for patients with EGFR-mutations, who exhibit disease progression despite EGFR-TKI treatment with MET-amplification. It can also be modified for inhibitors targeting other molecules.

Here, we present an assembloid model system to mimic tendon cellular crosstalk between the load-bearing tendon core tissue and an extrinsic compartment containing cell populations activated by disease and injury. As an important use case, we demonstrate how the system can be deployed to probe disease-relevant activation of extrinsic endothelial cells.

We present a protocol to investigate the crosstalk between X-ray-irradiated Caco-2 and peripheral blood mononuclear cells (PBMC). The protocol starts with Caco-2 irradiation and set-up of the co-culture with PBMC; subsequently, trans-epithelial electrical resistance is measured regularly over 48 h and western blot performed in both Caco-2 and PBMC.
