Ethidium Bromide

Ethidium bromide is a fluorescent intercalating dye that binds nucleic acids and has long been used to visualize DNA and RNA in biology. Its planar molecules insert between stacked base pairs, and binding increases fluorescence, allowing nucleic acids separated by agarose gel electrophoresis to appear under ultraviolet illumination. Researchers use this signal to estimate fragment size, assess sample integrity, and monitor amplification or restriction-digestion results. Because ethidium bromide can damage genetic material and requires careful handling and disposal, laboratories often apply strict safety procedures or select less hazardous fluorescent stains as alternatives.

Here, we present a protocol to use thiazole orange for the detection of DNA in gel electrophoresis experiments. The use of thiazole orange allows elimination of ethidium bromide, and fluorescence detection can be achieved with either UV or blue light.
