Excitation Secretion Coupling

Excitation-secretion coupling is the biological process that converts electrical activity in an excitable cell into the release of chemical messengers, linking cellular signaling to communication. When membrane depolarization reaches the cell surface, voltage-gated calcium channels open, allowing calcium ions to enter and activate synaptic or secretory vesicles for membrane fusion and exocytosis. This mechanism operates in neurons, neuroendocrine cells, and endocrine tissues, where it regulates neurotransmitter and hormone release. Studying excitation-secretion coupling helps explain synaptic transmission, hormone regulation, and disorders caused by altered calcium signaling or vesicle trafficking, while supporting the development of drugs that modify cellular secretion.

This paper describes a protocol that assesses the changes of myofilament Ca2+ sensitivity during contraction in isolated cardiac myocytes from rat heart. Together with cardiac electrophysiology, systolic/diastolic cytosol Ca2+ levels and contraction/relaxation, this measurement is imperative in underpinning the mechanisms mediating cardiac excitation-contraction coupling in healthy and diseased hearts.
