Fibroblasts

Fibroblasts are connective-tissue cells that produce and maintain the extracellular matrix, making them essential for tissue structure, repair, and homeostasis. They respond to biochemical and mechanical signals by synthesizing components such as collagen, fibronectin, and proteoglycans, while remodeling the matrix through regulated secretion and degradation. During wound healing, fibroblasts migrate into damaged tissue, proliferate, and support scar formation; persistent activation can contribute to pathological fibrosis. In biology and biomedical research, fibroblast cultures help investigators study cell signaling, tissue repair, extracellular-matrix organization, disease mechanisms, and the development of regenerative or anti-fibrotic therapies.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.

Traditional fibroblast research relies upon in vitro manipulation, masking in vivo fibroblast behavior. We describe a FACS-based protocol for the isolation of mouse skin fibroblasts that does not require cell culture.
