Fluorescence Spectrophotometer

A fluorescence spectrophotometer is an analytical instrument that measures light emitted by a fluorescent substance after it absorbs light at a selected wavelength, providing sensitive information about molecular composition and behavior. In operation, the instrument directs excitation light onto a sample, separates emitted light from scattered excitation light, and records fluorescence intensity across wavelengths to produce an emission spectrum. In biology, fluorescence spectrophotometers help quantify proteins, nucleic acids, metabolites, and fluorescent labels, while revealing changes in molecular environment, concentration, or interactions. These measurements support biochemical assays, enzyme studies, drug research, and investigations of cellular processes.

Microvolume samples are quantified by a spectrophotometer system that uses natural surface tension to retain samples without the use of cuvettes or capillaries. The dynamic range of protein concentrations and speed by which they can be measured are greatly increased with this method.

This communication presents data on the accuracy and reproducibility of the BioSpec-nano UV-VIS spectrophotometer for dsDNA and protein quantitation. Even with ultra-small volumes (1 to 2 L), reproducibility is excellent, while the automated wiping function improves throughput and results in minimal carryover for more precise results.
