Freeze Thaw Lysis

Freeze-thaw lysis is a physical method for breaking open biological cells to release intracellular contents, making it useful for sample preparation in biology. Repeated cycles of freezing and thawing cause ice crystals to form and expand inside cells, while associated osmotic and mechanical stresses disrupt the plasma membrane; thawing then permits cellular components to diffuse into the surrounding solution. The method can lyse cultured cells, bacteria, and some tissues without relying on chemical detergents, supporting the extraction of proteins, nucleic acids, enzymes, and organelles. Its simplicity and low reagent requirements make it valuable for laboratory research, although multiple cycles or additional disruption methods may be needed for resilient cell walls.
