Freezer Mill

A freezer mill is a laboratory instrument that mechanically pulverizes samples while they remain frozen, enabling efficient preparation of hard, fibrous, or biologically complex materials. The instrument repeatedly impacts or agitates a sealed container holding the sample and grinding media, while cryogenic cooling maintains low temperatures that limit heat generation, oxidation, and enzymatic degradation. In biology, freezer milling helps homogenize tissues, plant material, cells, and other specimens before extracting DNA, RNA, proteins, or metabolites. By producing a consistent, fine powder while preserving temperature-sensitive components, the method supports molecular analysis, biochemical assays, and reproducible sample preparation.

We describe a reliable method for the preparation of whole cell extracts from yeast or other cells using a cryogenic freezer mill that minimizes degradation and denaturation of proteins. The cell extracts are suitable for purification of functional protein complexes, proteomic analyses, co-immunoprecipitation studies and detection of labile protein modifications.

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.

Regulating Temperature in the Lab: Preserving Samples Using Cold

2023

Preservation of laboratory samples, specimens, and reagents using extreme cold is routinely performed in biomedical research labs. This video will discuss some of the methods for keeping laboratory samples cold and will explain the correct cooling method to use for each experimental requirement. For example, cooling agents, such as ice and dry ice, are typically used when keeping samples cold during experiments. This video discusses the physical properties of the most commonly used cooling...

Here, we present a protocol to demonstrate the generation of ice when water is introduced to a cold bath of brine, as a secondary refrigerant, at a range of temperatures well below the freezing point of water. It can be used as an alternative way of producing ice for industry.
