Frozen Core Decontamination

Frozen core decontamination is the controlled removal of surface contaminants from frozen biological or environmental core samples before analysis. Researchers maintain the sample in a frozen state while removing or isolating the outer layer, which may have contacted drilling equipment, handling surfaces, or the surrounding environment; sterile tools and containers help prevent recontamination. This preparation is especially important for ice-core studies of microorganisms, ancient DNA, and trapped chemical records. By separating the uncontaminated interior from introduced material, the method improves confidence that detected cells, molecules, or signals reflect the original frozen environment rather than laboratory or field contamination.

The cryosphere offers access to preserved organisms that persisted under past environmental conditions. A protocol is presented to collect and decontaminate permafrost cores of soils and ice. The absence of exogenous colonies and DNA suggest that microorganisms detected represent the material, rather than contamination from drilling or processing.
