Gc-ms Analysis

GC-MS analysis, or gas chromatography–mass spectrometry, is an analytical method that separates and identifies volatile compounds in complex samples, making it valuable for chemical characterization in biology. Gas chromatography first vaporizes the sample and separates its components as they travel through a column, while mass spectrometry ionizes each compound and measures the mass-to-charge ratios of the resulting ions to produce a characteristic spectrum. Researchers use GC-MS to analyze metabolites, lipids, environmental contaminants, drugs, and other biologically relevant molecules. Comparing spectra with reference libraries can support compound identification, while signal intensity provides information for qualitative and quantitative studies.

The sample is either a gas or a liquid that is vaporized in the injection port. Typically, the compounds analyzed are less than 1,000 Da, because it is difficult to vaporize larger compounds. GC is popular for environmental monitoring and industrial applications because it is very reliable and can be run nearly continuously.

Mutations in the kisspeptin receptor (KISS1R) are associated with reproductive disorders in patients. Here we describe how to introduce mutations of interest in the GC-rich sequence of KISS1R as well as the use of KISS1R constructs to characterize the degradation pathway of the receptor by immunoprecipitation and western blot.

GC-based Detection of Aldononitrile Acetate Derivatized Glucosamine and Muramic Acid for Microbial Residue Determination in Soil

We describe a method protocol for the GC-based analysis of the aldonitrile acetate derivatives of glucosamine and muramic acid extracted from soil. For elucidation of the chemical mechanism, we also present a strategy to confirm the structure of the derivative and the ion fragments formed upon electron ionization.

Minor workers of the ant species Colobopsis explodens are dissected to isolate the wax-like content stored in their hypertrophied mandibular gland reservoirs for subsequent solvent-extraction and analysis by gas chromatography-mass spectrometry. The annotation and identification of volatile constituents using the open-source software MetaboliteDetector is also described.
