Gel Washing Protocol

A gel washing protocol is a laboratory procedure for rinsing an electrophoresis gel to remove residual sample components, detergents, salts, or staining reagents before downstream analysis. Washing works through repeated exposure to a compatible buffer or solvent, allowing unwanted molecules to diffuse out of the gel while preserving separated proteins or nucleic acids within its polymer network. In biology, careful washing can improve staining contrast, reduce background, and prepare gels for imaging, transfer, enzymatic assays, or other detection methods. The choice of wash solution, duration, temperature, and number of exchanges depends on the gel composition and the intended application.
