Gene Amplification

Gene amplification is the production of multiple copies of a specific DNA sequence, a process that increases gene dosage and enables sensitive analysis of genetic material. In cells, it can arise through repeated DNA replication or rearrangements that duplicate genomic regions; in the laboratory, polymerase chain reaction (PCR) amplifies a target through cycles of denaturation, primer annealing, and DNA synthesis by a heat-stable polymerase. Amplified genes can alter protein production and contribute to biological phenomena such as tumor development, while controlled amplification provides abundant material for study. Researchers use gene amplification to detect pathogens, identify mutations, quantify nucleic acids, and support cloning, sequencing, and diagnostic testing.
