Gene Identification

Gene identification is the process of locating and characterizing a gene associated with a biological trait, function, or disease, making it central to genetics and modern biology. Researchers compare DNA sequences, genetic markers, inheritance patterns, and gene-expression profiles to narrow a genomic region and determine which gene or variant contributes to the observed phenotype. Methods such as genome-wide association studies, linkage analysis, DNA sequencing, and functional validation help distinguish causal changes from harmless variation. Identifying genes supports disease diagnosis, evolutionary studies, targeted drug development, and research into cellular processes, while providing a foundation for personalized medicine and genetic engineering.

VIGS-Mediated Forward Genetics Screening for Identification of Genes Involved in Nonhost Resistance

Virus-induced gene silencing is an useful tool for identifying genes involved in nonhost resistance of plants. We demonstrate the use of bacterial pathogens expressing GFPuv in identifying gene silenced plants susceptible to nonhost pathogens. This approach is easy, fast and facilitates large scale screening and similar protocol can be applied to studying various other plant-microbe interactions.

Matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) and molecular techniques (16S rRNA gene sequencing) permit the identification of rare bacterial pathogens in routine diagnostics. The goal of this protocol lies in the combination of both techniques which leads to more accurate and reliable data.

This paper describes two phenotyping methods without the use of epidermal peels to characterize the genes controlling stomatal development. The first method demonstrates how to analyze a stomatal phenotype using a toluidine blue O-stained plant epidermis. The second method describes how to identify stomatal ligands and monitor their biological activities.
