Gfp Tubulin

GFP-tubulin is a fluorescent fusion protein that labels microtubules, the dynamic cytoskeletal polymers that organize cell shape, intracellular transport, and chromosome movement. It combines tubulin with green fluorescent protein (GFP), allowing the labeled tubulin to incorporate into microtubules while GFP emits visible fluorescence when excited by appropriate light. In biology, GFP-tubulin enables live-cell imaging of microtubule assembly and disassembly, organization of the mitotic spindle, and changes in cytoskeletal architecture over time. This approach helps researchers connect microtubule dynamics with cell division, migration, intracellular trafficking, and responses to drugs or genetic perturbations.

This protocol describes a technique for the high-yield isolation of tubulin from the porcine brain optimized for small-scale instrumentation. The isolation procedures are complemented by procedures for determining tubulin polymerization activity in vitro using co-sedimentation assays and transmission electron microscopy.

While the transport of cell surface proteins is relatively easily studied, visualizing the trafficking of intracellular proteins is much more difficult. Here, we use constructs incorporating photoactivatable GFP and demonstrate a method to accurately follow the amyloid precursor protein from the Golgi apparatus to down-stream compartments and follow its clearance.
