Glass Bead Spreading

Glass bead spreading is a microbiological technique for distributing a liquid cell suspension evenly across the surface of solid growth medium, enabling researchers to isolate and quantify microorganisms. In this method, a measured inoculum is placed on agar with sterile glass beads, and gentle agitation rolls the beads across the surface, dispersing cells into separate locations as the liquid is absorbed. The resulting spatial separation supports colony formation, viable cell counting, and selection of individual colonies for further analysis. In biology, glass bead spreading provides a simple, reproducible approach for culturing bacteria, screening phenotypes, and assessing microbial growth under defined experimental conditions.

A method for surface-spreading chromosomes from budding yeast is presented. This method is derived from a method previously described by Loidl and Klein. In addition, we demonstrate a procedure for immunostaining of spread chromosomes.
