Hematopoietic Reconstitution

Hematopoietic reconstitution is the restoration of the blood-forming system after depletion or transplantation, a process essential for recovering oxygen transport, immune defense, and blood clotting. It occurs when hematopoietic stem and progenitor cells establish themselves in the bone marrow, self-renew, and differentiate into red blood cells, immune cells, and platelets. In biology and medicine, studying hematopoietic reconstitution helps assess stem cell engraftment, bone marrow transplantation, immune-system recovery, and the effects of chemotherapy or radiation. These insights also support research into graft compatibility, blood disorders, and cell-based therapies.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Efforts to isolate the catalytic subunit of telomerase, TERT, in sufficient quantities for structural studies, have been met with limited success for more than a decade. Here, we present methods for the isolation of the recombinant Tribolium castaneum TERT (TcTERT) and the reconstitution of the active T. castaneum telomerase ribonucleoprotein (RNP) complex in vitro.

A method to analyse the distribution of bone marrow hematopoietic progenitors in flow cytometry as well as to efficiently isolate highly purified hematopoietic stem cells (HSCs) is described. The isolation procedure is essentially based on magnetic enrichment of c-Kit+ cells and cell sorting to purify HSCs for cellular and molecular studies.
