Heme Extraction

Heme extraction is the process of isolating heme, an iron-containing porphyrin group found in hemoglobin and other biological proteins, for biochemical analysis. The procedure typically disrupts cells or proteins, denatures the globin component, and uses acidic conditions or organic solvents to separate the relatively hydrophobic heme from surrounding biomolecules; subsequent purification can preserve its characteristic color and iron center. In biology, extracted heme supports measurements of heme content, studies of hemoglobin structure and turnover, investigations of heme-binding proteins, and analyses of pathways involved in oxygen transport, cellular respiration, and porphyrin metabolism.

Altered intracellular heme levels are associated with common diseases such as cancer. Thus, there is a need to measure heme biosynthesis levels in diverse cells. The goal of this protocol is to provide a fast and sensitive method to measure and compare the levels of heme synthesis in different cells.

Bathophenanthroline Sulfonate-Based Colorimetric Assay: A Simple and Rapid Method for Quantitation of Non-Heme Iron in Mouse Liver Tissue

In this video, we describe a colorimetric-based method to determine non-heme iron content in mouse liver tissue. Bathophenanthroline sulfonate is used as the chromogenic substrate in the presence of a reducing agent, which converts ferric iron to chelatable ferrous iron.

Bioluminescence Imaging of Heme Oxygenase-1 Upregulation in the Gua Sha Procedure

Gua Sha, traditional Chinese therapeutic skin scraping, causes subcutaneous microvascular blood extravasation. We report a protocol of bioluminescence imaging of HO-1-luciferase transgenic mice to demonstrate that Gua Sha upregulates heme oxygenase-1 (HO-1) in multiple organs.
