Hepatocyte Death

Hepatocyte death is the loss of liver parenchymal cells through regulated or uncontrolled processes, and it is central to liver development, tissue maintenance, disease, and repair. It can occur through apoptosis, in which caspase activation produces controlled cellular dismantling, or necrosis, in which severe injury disrupts membranes, releases intracellular contents, and promotes inflammation. Studying these pathways helps explain liver damage caused by toxins, infections, metabolic disorders, and impaired blood flow. Methods for measuring hepatocyte death support disease modeling, evaluation of liver regeneration, and safety testing for pharmaceuticals and other chemical exposures.

To help assess the molecular mechanisms underlying zebrafish biliary-driven liver regeneration, we established a liver injury model in which the nitroreductase-expressing hepatocytes are genetically ablated upon metronidazole treatment. In this protocol, we describe how to adeptly manipulate, monitor and analyze hepatocyte ablation and biliary-driven liver regeneration.

De novo lipogenesis and β-fatty acid oxidation constitute key metabolic pathways in hepatocyte, pathways that are perturbed in several metabolic disorders, including fatty liver disease. Here we demonstrate isolation of mouse primary hepatocytes and describe quantification of β-fatty acid oxidation and lipogenesis.

Isolation of Human Hepatocytes by a Two-step Collagenase Perfusion Procedure

A modified two-step collagenase perfusion procedure for isolation of human hepatocytes is described. This method can also be applied to other mammalian livers. The isolated hepatocytes are available in high yield and viability, making them a suitable model for scientific research in areas such as liver regeneration, pharmacokinetics and toxicology.
