Hif1 Beta Subunit

The HIF-1 beta subunit, also called aryl hydrocarbon receptor nuclear translocator (ARNT), is a constitutively expressed transcription factor that helps cells respond to changes in oxygen availability. It contains basic helix-loop-helix and PAS domains that enable it to dimerize with HIF-1 alpha; under hypoxic conditions, this complex binds hypoxia-response elements in DNA and activates specific genes. HIF-1 beta therefore contributes to oxygen homeostasis by regulating processes such as angiogenesis, glucose metabolism, and cellular adaptation to low oxygen. Studying this subunit is important for understanding development, cancer biology, ischemic disease, and other conditions involving disrupted oxygen sensing.

This publication presents a protocol showing how to measure the activity of recombinant protein phosphatase 2A catalytic subunit (PP2Ac) in response to recombinant α-synuclein, β-synuclein, or γ-synuclein proteins using a simple colorimetric assay. We show findings regarding α-synuclein specificity toward PP2Ac in mouse brain.

Here, we describe a detailed protocol for the in vitro reassociation of purified, salt-washed eukaryotic ribosomal subunits for the analysis of 80S particle formation.
