Histone Acetylation

Histone acetylation is a reversible epigenetic modification in which acetyl groups are added to lysine residues on histone proteins, helping regulate access to DNA. Histone acetyltransferases add these groups and reduce the positive charge of histones, weakening histone-DNA interactions and promoting a more open chromatin structure; histone deacetylases remove them and can restore tighter packing. By influencing transcription without changing the DNA sequence, this process contributes to cell differentiation, development, and responses to environmental signals, while abnormal acetylation patterns are associated with disease and are investigated as targets for therapeutic intervention.

This manuscript describes an experimental approach to morphologically and biochemically characterize horse oocytes. Specifically, the present work illustrates how to collect immature and mature horse oocytes by ultrasound-guided ovum pick-up (OPU) and how to investigate chromosome segregation, spindle morphology, global histone acetylation, and mRNA expression.
