Humane Endpoints

Humane endpoints are predefined criteria and actions used to prevent or minimize unnecessary pain, distress, or suffering in animals involved in biological research. Researchers monitor indicators such as changes in behavior, body condition, mobility, feeding, and clinical status, then intervene when an animal reaches a specified threshold, often through supportive care, removal from a study, or euthanasia. Establishing humane endpoints strengthens animal welfare, improves consistency in experimental decisions, and supports responsible study design by balancing scientific objectives with ethical obligations. Clear monitoring plans and trained personnel help identify deterioration early and reduce avoidable suffering.

In argentometric precipitation titrations, endpoints can be detected visually by the Mohr, Volhard, and Fajans methods. Here, we present a protocol to establish important endpoints and proliferative markers of small intestinal injury and compensatory hyperproliferation using a model of chemotherapy-induced mucositis. We demonstrate the detection of proliferating cells using a cell cycle specific marker and using small intestinal weight, crypt depth, and villus height as endpoints.

Investigation of the Uncanny Valley Hypothesis and affective experience requires an understanding of the hypothesis' dimension of human likeness (DHL). This protocol allows representation of the DHL and examination of categorical perception. Use of the same stimuli and fMRI to distinguish brain regions responsive to physical and category change is illustrated.
