Immunofluorescent Localization

Immunofluorescent localization is a microscopy-based technique that identifies where specific proteins or cellular structures occur within fixed cells or tissues, linking molecular identity to spatial organization. The method uses antibodies to bind target molecules, with fluorescent labels attached directly to primary antibodies or to secondary antibodies; illumination at an appropriate excitation wavelength causes the labels to emit detectable light, producing localized signals. Researchers use these patterns to assess protein distribution, co-localization, cell architecture, and changes associated with development, infection, disease, or experimental treatment, making the technique valuable for connecting molecular processes with cellular function.

An In Vivo Immunofluorescence Localization Method to Study Antibody Biodistribution

This video demonstrates an in vivo immunofluorescence localization (IVIL) technique to study the biodistribution of tumor-targeting antibodies in a murine model of breast cancer. Upon the intravenous injection of a tumor-specific antibody in the mouse, the tumor is surgically isolated, and the localization of the antibody within the tumor tissue is confirmed via ex vivo immunostaining.

Localization and Quantification of Begomoviruses in Whitefly Tissues by Immunofluorescence and Quantitative PCR

We describe an immunofluorescence and quantitative PCR method for the localization and quantification of begomoviruses in insect tissues. The immunofluorescence protocol can be used to colocalize viral and vector proteins. The quantitative PCR protocol can be extended to quantify viruses in whole whitefly bodies and virus-infected plants.
