Infection Quantification

Infection quantification is the measurement of the amount, prevalence, or activity of an infectious agent in a biological sample, providing a basis for evaluating disease progression and treatment response. Depending on the organism and research question, investigators estimate infection by counting viable colonies, detecting pathogen-specific nucleic acids or antigens, or measuring infected cells and associated biological signals; each approach captures a different aspect of infection. These measurements support studies of host-pathogen interactions, antimicrobial efficacy, transmission, and disease mechanisms, while helping researchers compare experimental conditions and assess how infection changes over time.

Establishment and Quantification of De Novo Lytic Infection by Cell-free Kaposi's Sarcoma-Associated Herpesvirus

2025

This protocol describes an in vitro system to model de novo lytic infection of Kaposi's sarcoma-associated herpesvirus using BAC16-derived virions. The method enables investigation of early viral replication and dissemination. Infectious virus production is quantified by GFP-based infectious units assays and real-time PCR of encapsidated viral genomes.

Quantification of Coronavirus Infection Using a Dual-Reporter Assay

Take a culture of genetically modified epithelial cells expressing two reporter proteins: a constitutively active Renilla luciferase or Rluc, which acts as a control, and a firefly luciferase or FFluc, rendered inactive by insertion of a protease-cleavable oligopeptide.Infect the cells with a coronavirus. The virus releases its RNA and expresses the viral protease, which cleaves the oligopeptide, activating FFluc.Replace the media with a buffer containing a detergent and the FFluc substrate.The...

Quantification of Helicobacter pylori Load in an Infected Mouse Stomach

2025

In this video, we describe a protocol to quantify Helicobacter pylori load in mouse stomach tissues post-infection to confirm its colonization.

Quantification of Inflammatory Mediators in Infected Human Tissue-Derived Cells

2025

This video demonstrates the quantification of inflammatory mediators during infection by incubating human lung-tissue-derived immune cells with Haemophilus influenzae. Flow cytometry is used to measure cytokine fluorescence in labeled lymphocytes, correlating with the production of pro-inflammatory cytokines in response to bacterial infection.

Quantification of Bacterial Load in Mouse Spleen and Liver After Systemic Infection

Source: Ahn, J. J., et al. Experimental Infection with Listeria monocytogenes as a Model for Studying Host Interferon-γ Responses. J. Vis. Exp. (2016)The video demonstrates bacterial load measurement in spleen and liver tissues isolated from mice infected with an intracellular bacterial pathogen. Bacteria, accumulated within tissue-resident phagocytes during systemic infection, are released using detergent-based lysis and mechanical disruption. Serial dilutions of the homogenates are plated on...
