Lentivirus Overexpression

Lentivirus overexpression is a gene delivery method that uses engineered lentiviral vectors to produce unusually high levels of a selected gene in target cells. The vector carries an expression cassette into cells, where viral entry enables delivery of the genetic material and, in many systems, stable integration into the host genome, allowing sustained transcription of the introduced gene. In biology research, this approach supports functional studies by increasing protein production in cultured cells, helping investigators assess gene function, signaling pathways, cellular phenotypes, and responses to experimental treatments. Its ability to provide long-term expression makes it useful for studying genes that are difficult to introduce through transient transfection.

MicroRNAs have significant roles in brain structure and function. Here we describe a method to enforce hippocampal miRNA over-expression using stereotactic injection of an engineered miRNA-expressing lentivirus. This approach can serve as a relatively rapid way to assess the in vivo effects of over-expressed miRNAs in specific brain regions.
