Inflammatory Mediator Measurement

Inflammatory mediator measurement is the quantitative assessment of signaling molecules that regulate immune and inflammatory responses, including cytokines, chemokines, and lipid mediators. Researchers typically analyze biological samples such as blood, tissue extracts, or cell-culture media using antibody-based assays, multiplex platforms, or mass spectrometry, in which molecular recognition produces a signal proportional to mediator concentration. We describe a simple immunoassay to measure the production of pro-inflammatory cytokines, such as IL-1 beta production, in patients presenting with autoinflammatory phenotypes. By activating cells in whole blood cultures with pathogen-associated molecular patterns, specifically with lipopolysaccharide, cytokine secretion can be conveniently evaluated in whole blood supernatants.
