Intact Method

The intact method is a biological analysis approach that characterizes proteins or other biomolecules in their complete, undigested form, preserving information that fragmentation can obscure. In intact protein analysis, a purified sample is introduced into a mass spectrometer, where ionization produces charged molecular species and the instrument measures their mass-to-charge ratios to determine molecular mass and detect changes such as post-translational modifications. This strategy supports protein identity confirmation, assessment of sample heterogeneity, and quality control in biochemistry and biopharmaceutical research. By retaining the whole molecule, the method complements peptide-based workflows and can provide a direct view of molecular composition.

This paper describes a methodology to prepare cardiovascular tissue samples for MS analysis that allows for (1) the analysis of ECM protein composition, (2) the identification of glycosylation sites, and (3) the compositional characterization of glycan forms. This methodology can be applied, with minor modifications, to the study of the ECM in other tissues.

The main focus of this protocol is to efficiently isolate viable primary glomeruli cultures with minimal contaminants for use in a variety of downstream applications. The isolated glomeruli retain structural relationships between component cell types and can be cultured ex vivo for a short time.
