Interatrial Septum

The interatrial septum is the wall of tissue separating the right and left atria of the heart, where it helps organize blood flow and prevents mixing after birth. During embryonic development, coordinated growth of the septum primum and septum secundum forms an opening called the foramen ovale, which allows blood to bypass the fetal lungs; rising left atrial pressure after birth functionally closes this passage. Studying the interatrial septum helps explain normal cardiac development and conditions such as atrial septal defects and patent foramen ovale, supporting echocardiographic diagnosis, risk assessment, and congenital heart research.

Here, we describe an improvement of the semi-in vitro (SIV) method for observing pollen tube guidance and reception in Arabidopsis thaliana, which increases the receptivity of ovules. The high-throughput SIV cum septum method may be coupled with gametophyte marker lines and genetically encoded biosensors to monitor the dynamic process of fertilization.

Sinoatrial Node Isolation from Mouse Model: A Procedure to Harvest SA Node from Murine Heart

2025

This video demonstrates the procedure to isolate sinoatrial node or SAN from mouse heart. The SAN houses the sinoatrial cells that initiates heartbeat by generating spontaneous action potentials.

Catheter-based Endovascular Angioplasty for Fibrosing Mediastinitis-associated Pulmonary Vein Stenosis

2025

Pulmonary vein stenosis (PVS) is a life-threatening complication of fibrosing mediastinitis (FM), with a poor prognosis if not properly treated. Currently, catheter-based endovascular angioplasty is a promising therapeutic strategy for FM-associated PVS. This protocol primarily outlines a stepwise approach to the transcatheter procedure for FM-associated PVS.

Whole Mount Labeling of Cilia in the Main Olfactory System of Mice

Cilia of olfactory sensory neurons contain proteins of the signal transduction cascade, but a detailed spatial analysis of their distribution is difficult in cryosections. We describe here an optimized approach for whole mount labeling and en face visualization of ciliary proteins.

Perforated Patch-Clamp Recording of Olfactory Sensory Neurons in Mice

2025

This video demonstrates electrophysiological recording of olfactory sensory neurons (OSNs) in mice using a perforated patch-clamp technique. The olfactory epithelium, containing OSNs with fluorophore-tagged odor receptors, is isolated from a mouse and secured in a recording chamber. A recording pipette is clamped to the dendritic knob of an OSN, and nystatin within the pipette perforates the membrane to form electrical connectivity with the cell. Finally, another pipette is used to deliver an...
