Intestinal Permeability

Intestinal permeability is the selective ability of the intestinal lining to regulate movement between the gut lumen and bloodstream, allowing nutrients to enter while limiting pathogens and harmful compounds. This barrier depends on mucus, epithelial cells, membrane transporters, and tight junctions that control paracellular passage between cells, while transcellular pathways permit specific molecules to cross through the cells. In biology and biomedical research, measuring intestinal permeability helps assess barrier integrity, investigate inflammation and infection, and evaluate how diet, drugs, or disease alter gut function. These studies clarify how epithelial barrier disruption may influence local and systemic physiology.

(2019)This video demonstrates a fluorescence-based assay to quantify intestinal permeability in C. elegans exposed to Pseudomonas aeruginosa. Compared to controls fed Escherichia coli, pathogen-treated worms show increased fluorophore leakage into the body cavity, indicating epithelial barrier damage.

This protocol describes the use of excised intestinal tissue preparations or "intestinal sacs" as an ex vivo model of intestinal barrier function. This model may be used to assess integrity of both the epithelial barrier and the mucous gel layer at specific intestinal sites in animal models of digestive disease.
