Lens Capsule Isolation

Lens capsule isolation is a biological technique that separates the thin, elastic basement membrane surrounding the vertebrate eye lens from the lens fibers and associated tissue. Under microscopic observation, researchers dissect the lens and carefully remove its cellular contents while preserving the capsule, which can retain adherent lens epithelial cells. The isolated capsule provides a model for studying lens development, epithelial cell behavior, wound healing, and cataract-associated changes. It also supports analyses of capsule mechanics and cell–matrix interactions, helping connect structural properties of the lens to processes involved in vision and ocular disease.

RNA Isolation from Mouse Ocular Lens Epithelium and Fiber Cell Bulk Masses

2025

This protocol details the separation of the mouse ocular lens epithelium and fiber cell bulk mass, followed by RNA isolation and qPCR analysis. This method allows for separating lens cell compartments for a more detailed analysis of the transcriptome and biological processes in epithelial cells vs. differentiated fiber cells.

Culturing and Isolating Capsule-Forming Pathogenic Bacterial Strains

(2019)This video demonstrates the preparation and characterization of bacterial strains with varying capsule production. Pathogenic strains carrying different mutations in capsule-related genes are cultured, grown in liquid media, and subjected to centrifugation to distinguish capsule phenotypes based on pellet density. Buffer treatment is used to wash and stabilize the cells...

DiRita1 1 Department of Microbiology and Molecular Genetics, Michigan State University, East Lansing, Michigan, United States of America Bacteria are diverse microorganisms found nearly everywhere on Earth. Many properties help distinguish them from each other, including but not limited to Gram-staining type, shape and arrangement, production of capsule, and formation of spores. To observe these properties, one...

Human pluripotent stem cells (hPSCs) have the potential to treat a myriad of different diseases. The utility of these cells lies in the fact that they can differentiate into any cell type in the body. Here we describe the teratoma assay, which is used to demonstrate the pluripotence of hPSCs.
