Light Dependent Reactions

Light-dependent reactions are the photosynthetic processes that convert light energy into chemical energy, making them essential for plant, algal, and cyanobacterial growth. In chloroplast thylakoid membranes, chlorophyll absorbs photons and energizes electrons; water is split to replace lost electrons, releasing oxygen, while electron transport drives proton movement and ATP synthase produces ATP, and NADP+ is reduced to NADPH. ATP and NADPH then fuel carbon fixation in the Calvin cycle. Understanding these reactions clarifies how photosynthetic organisms capture solar energy, release atmospheric oxygen, and support applications in plant physiology, crop improvement, and bioenergy research.

We describe the use of a stopped-flow instrument to investigate both the reductive and oxidative half-reactions of Aspergillus fumigatus siderophore A (SidA), a flavin-dependent monooxygenase. We then show the spectra corresponding to the species in the reaction of SidA and we calculate the rate constants for their formation.
