Lipogenesis

Lipogenesis is the biological process of synthesizing fatty acids and storing excess energy as triglycerides, helping organisms regulate fuel availability and energy balance. When carbohydrate or other nutrient supply exceeds immediate demand, cells convert acetyl-CoA into malonyl-CoA through acetyl-CoA carboxylase, then fatty acid synthase builds fatty acid chains that can be esterified into triglycerides. Insulin and nutrient signals promote this pathway, particularly in the liver and adipose tissue. Studying lipogenesis clarifies how metabolism responds to diet and energy status, while its dysregulation contributes to obesity, fatty liver disease, insulin resistance, and other metabolic disorders.

De novo lipogenesis and β-fatty acid oxidation constitute key metabolic pathways in hepatocyte, pathways that are perturbed in several metabolic disorders, including fatty liver disease. Here we demonstrate isolation of mouse primary hepatocytes and describe quantification of β-fatty acid oxidation and lipogenesis.
