Lung Tissue Processing

Lung tissue processing is the preparation of lung specimens for microscopic, molecular, or histological analysis, making it possible to examine tissue structure, cell types, and disease-related changes. The workflow typically preserves, fixes, and sections tissue so that its architecture and biomolecules remain suitable for staining, imaging, or downstream assays; processing conditions must be selected to limit distortion and degradation. In biology, these methods support studies of respiratory development, infection, inflammation, fibrosis, cancer, and toxicology. Consistent processing improves the quality and comparability of experimental results, helping researchers connect cellular and structural findings with lung function and disease mechanisms.

The stiffness of the extracellular matrix strongly influences multiple behaviors of adherent cells. Matrix stiffness varies spatially throughout a tissue, and undergoes modification in various disease conditions. Here we develop methods to characterize spatial variations in stiffness in normal and fibrotic mouse lung tissue using atomic force microscopy microindentation.
